The sample preparation systems can be integrated with any existing separation and detection system. A fully automated SPE cartridge system exists of:

Conditioning

While the previous SPE cartridge is still being eluted to the separation column. A new SPE cartridge is placed in the other clamp. Solvent lines and the new SPE cartridge are flushed with appropriate solvents to prepare for the next sample introduction.

Sample loading

Sample is aspirated from the vial or well into the sample loop

Extraction

Sample is flushed from the sample loop through the SPE cartridge. Analytes are trapped and concentrated, most of the matrix is flushed to waste

Wash

Additional volumes of solvent are flushed through the SPE cartridge to remove more matrix components

Elution

Extracted and concentrated analytes are eluted from the cartridge onto the analytical separation column with the HPLC mobile phase

The ROXY™ EC is the ideal introduction system for Electrospray Ionization (ESI)-MS for single compound conversion. For known compounds (e.g., drug candidates, pollutants, pesticides, etc) a quick profiling and/or reconfirmation of potential metabolites is often sufficient for decision making. Therefore, target compounds can be directly infused through the electrochemical ReactorCell™ into the MS, often without the need of any liquid chromatographic separation (HPLC), making EC/MS a very powerful tool for fast metabolic profiling and related applications.

HPLC/ECD in Neuroscience

Unlike many other detectors for HPLC, amperometric electrochemical detection (ECD) can be miniaturized without losing detection sensitivity. Generally accepted benefits of micro HPLC are the improvement in performance (less peak dilution) and the reduced solvent consumption. But most interesting feature of miniaturized HPLC-ECD is the ability to analyze small samples while maintaining an excellent signal to noise ratio (S/N), as both signal and noise decrease with miniaturized electrodes. In fact the optimum S/N is obtained for amperometric cells with electrodes in the range of 0.5 – 5 mm ID. We found the best detection sensitivity is obtained when electrode diameter matches the column ID. It is this feature that made HPLC with electrochemical detection the method of choice for neurotransmitter analysis, especially in combination with microdialysis sampling.

Microdialysis

In vivo microdialysis has become an invaluable tool that provides real-time information of neurotransmitter untitled-11levels in living brain. Microdialysis samples are collected and stored, or analysed immediately in an on-line configuration using HPLC-ECD. It is an analytical challenge to provide reproducible and accurate data as neurotransmitter levels are often below the nanomolar and sometimes even below picomolar concentration range.

ALEXYS Neurotransmitter Analyzers

The ALEXYS product line comprises a number of integrated system solutions which have been developed for trace analysis of neurotransmitters such as noradrenaline, dopamine, serotonin and metabolites. Parallel and serial detection schemes using multiple flow cells have been developed. In stead of running sequential trials for different neurotransmitters it is now possible to do multi component analysis in a parallel HPLC configuration. Dual or triple loop injection valves are applied with minimum sample consumption on parallel HPLC systems under completely different conditions. Getting more information out of less sample is not only saving time and money but – in the end – also rodents.

XPLORER AOX/TOX/POX/EOX

Elemental Combustion Analyzer for Adsorbable Organic Halogens (AOX), TOX, EOX and POX by Microcoulometry

The TE Instruments XPLORER is an AOX/TOX, POX and EOX combustion analyzer offering fast, accurate and untitled-7precise analysis of organic halogens. The AOX, EOX and POX modules are easily interchangeable. Assisted by a unique cup slider principle, the sample cup simply utilizes Newton’s law of gravity, allowing the cup or frit to ‘fall’ into the boat, before entering the horizontal furnace. When the boat returns form the furnace, the slider moves to the eject position, where the ‘burned’ cup or frit is collected for re-use, without any pile-up in the furnace tube. The newest technologies combined with years of experience created this unique combustion analyzer.
